CJC-1295 is a powerful synthetic analogue of growth hormone-releasing hormone (GHRH), composed of 30 amino acids. There are two variants of CJC-1295:

The first, GHRH, is produced in the hypothalamus and releases growth hormone in a pulsatile manner from the pituitary but has a very short half-life of just a few minutes. The active segment of GHRH, made up of the first 29 amino acids, is available as a manufactured peptide called Sermorelin. This was modified to extend its half-life to approximately 30 minutes, leading to the creation of CJC-1295. Further enhancements include the addition of DAC (Drug Affinity Complex), which binds to the protein albumin, extending its half-life to around 8 days, known as CJC-1295 + DAC. A non-DAC version is also available, creating a more natural growth hormone spike each night.

The longer half-life allows for just one or two injections per week, providing sustained stimulation for growth hormone release from the pituitary through the GHRH receptor, albeit this may reduce the amplitude of GH pulses and lead to decreased stimulation of GH-target tissues.

Safety Considerations:

To ensure optimal results, it's recommended to take 'hormone holidays' every three to six months while using long-acting forms of CJC. This allows the pituitary to recover, during which Sermorelin can serve as a substitute. These breaks may also help mitigate the risk of GH resistance, which can arise from antibodies that deactivate GH or a decrease in GH receptors on tissues (down-regulation). While these risks are theoretical, no long-term studies have definitively confirmed them.
